Hello. I have a Denon MC6000 Mk2 and I'm running virtual 2021. Everything works great except I would like to have volume control of my sampler from the mixer. Is there any way to set either channel 3 or 4 to play the sampler out?

Can do that in software, click the small dot at upper right corner in sampler in sideview.
And there you can set auto routing to deck 3 or 4
